The ability to be able to identify or know the steps to identify an unknown microorganism illustrates an intellectual and physical discipline. It also reflects the ability of one’s accumulation of knowledge and skills when able to proceed in identifying unknown bacteria. In microbiology class, students were given a tube of mixed cultures that needed to be separated to proceed in the identification of just one of the cultures. Through the course of a few weeks, materials and methods took place to lead to the final result. This will be explained in the following.
MATERIALS AND METHODS
Amongst the different unknowns given to groups of two, an unknown labeled three was given out via the lab instructor. It was told that the tube contained two cultures. With that being said, to be able to identify the two cultures separately the quadrant streak method was applied (Brown, 2007). The quadrant streak method involves a petri plate, loop, Bunsen burner and the skill of being able to properly streak the plate so cultures can separate and become pure. In order for the performance of the quadrant streak method B to be successful, the skill of proper aseptic technique was applied (Brown, 2007).
The aseptic technique was applied in all methods and procedures of the lab. Proper aseptic technique relies on the ability to inoculate a loop, flame mouth of tube, and properly insert organism within the medium of desired growth. In this case the medium was trypticase soy agar. The thorough method that was practiced can be found in the eleventh edition of the course laboratory manual by Alfred E. Brown (2009).

To conclude, the unknown project allowed the education learned in previous classes to be used for the steps in identifying an unknown bacterium. This project was a great opportunity to strengthen skills in research and understanding. Even though the final result may not be what’s lined up on the lab instructor’s chart. There is great knowledge and understanding attained from the experience.
