Introduction:
This lab focuses on the isolation and characterization of an unknown organisms expressing interesting properties in relation to Antimicrobial Activity. At the conclusion of this laboratory experience The researcher would be required to describe the isolates obtained from the soil macroscopic and microscopic appearance, Perform the gram stain of the microorganisms in question and to discriminate the organisms from other microbes that could be contained in the soil and to finally make certain that the organisms have not been exposed to outside sources and be isolated in a pure culture. The researcher must first isolate the microbe and try to grow the organism in a pure culture to commence with testing the organism. The isolation of Microbes in a laboratory and clinical settings are of the up most importance and due to the isolation of different microbes from each other various procedures become open for the researcher to use like six fold dilutions that allow for the quarantined microbe to be reduced down in number to ensure that the test are being run are being tested on just a single type of organism. This could be accounted for by stating that there was a very large amount of microbes present in the soil and the dilutions did not dilute enough of the microorganism out or human error could also account for Not having any statistically significant plates to use.
The bacterium that I replated was obtained from the YPD 10-3 media and it gave rise to bacterium 1 & 3, The other bacterium that I obtained came from LB 10-5 . The two bacterium from the YPD plate varied in color the bacterium that I refer to as one had a reddish color had a circular form a raised elevation and a entire margin, The bacterium that I refer to as 3 Had a grey appearance a circular form a raised elevation and the margin appeared to be entire. The bacterium that I obtained from the LB media had a white appearance had a circular form a faintly raised elevation and the margin was entire.

I identified the genus and species of an unknown bacterial culture, #16, and I applied the following knowledge of morphologic, cultural and metabolic characteristics of the unknown microorganism according to the laboratory manual as well as my class notes and power point print outs. I was given an incubated agar slant labeled #16 and a rack of different tests to either examine or perform myself; the tests are as follows: Gram Stain; Nutrient Gelatin Test; Carbohydrate Fermentation; Dextrose, Lactose and Sucrose; IMVIC tests; Citrate, Indole, Mythel-Red and Vogues Proskauer test; as well as a Urease and TSI Test.
After 48 hours of incubation the agar plates were viewed. Individual colonies were tested for successful isolation by gram staining and then viewing the stained bacteria under a microscope. Isolation was successful. One colony of each unknown bacteria was transferred to an agar slant for growth. The agar slants were stored at room temperature over the weekend so that they would not grow too much.
The purpose of this study is to identify an unknown bacterium from a mixed culture, by conducting different biochemical tests. Bacteria are an integral part of our ecosystem. They can be found anywhere and identifying them becomes crucial to understanding their characteristics and their effects on other living things, especially humans. Biochemical testing helps us identify the microorganism present with great accuracy. The tests used in this experiment are rudimentary but are fundamental starting points for tests used in medical labs and helps students attain a better understanding of how tests are conducted in a real lab setting. The first step in this process is to use gram-staining technique to narrow down the unknown bacteria into one of the two big domains; gram-negative and gram-positive. Once the gram type is identified, biochemical tests are conducted to narrow down the specific bacterial species. These biochemical tests are process of elimination that relies on the bacteria’s ability to breakdown certain kinds of food sources, their respiratory abilities and other biochemical conditions found in nature.
The integumentary system has five main functions. The five main functions of the integumentary system are protection, regulation, sensation, absorption, and secretion. It consists of the hair, nails, skin, sebaceous and sweat glands. The largest organ of the entire body is the skin. The skin consists of three different layers. The epidermis, the dermis, and the subcutaneous layer, these are the three layers of the skin. There is a layer that is deeper than the skin and contains body fat, that layer is called the hypodermis. Sweat glands are also called sudoriferous glands. One of the functions of the skin is to maintain homeostasis. Synthesis of vitamin D, excretion of urea, excretion of salts and water are a few things that the skin is also capable of. The top layer of the skin is called the epidermis. The epidermis contains five different layers. These layers are the stratum corneum, the stratum lucidum, the stratum spinosum, and the stratum basale. In the epidermis there are four different cell types. The keratinocytes which produce keratin, the merkel cells that are sensitive to touch, the langerhans cells which is a dendritic type off cell, and the melanocytes which produce melanin.
War always seems to have no end. A war between countries can cross the world, whether it is considered a world war or not. No one can be saved from the reaches of a violent war, not even those locked in a safe haven. War looms over all who recognize it. For some, knowing the war will be their future provides a reason for living, but for others the war represents the snatching of their lives without their consent. The integumentary system is composed of the skin and the structures related to the skin, which include the hair, the finger nails, the sensory receptors and the glands (Shier, Butler, & Lewis, 2009). This system has an important function since it provides protection to the body, helps to maintain body temperature, and contains sensory receptors (p.117). The skin has essential function such as regulating homeostasis and body temperature, also delaying the loss of water from deeper tissues, storing sensory receptor, synthesizing biomechanical, and discharging waste from the body (p.117). The skin has two layers: the epidermis (outer layer) and the dermis (inner layer) (p.117). There is also a subcutaneous layer or hypodermis (p.117). According to Shier, Butler, and Lewis 2009 “as its name indicates, this layer is beneath the skin and not a true layer of the skin” (p.117). The physical effects of war overwhelm the naïve causing pain and suffering. Initially, war entangles the lives of youth, destroying the innocence that they experience as an aspect of their life. The girl “glid[ing] gracefully down the path” (1) and the boy “rid[ing] eagerly down the road” (9) have their enjoyable realities striped by the harshness of war. Likewise, war enters women’s lives creating turmoil. The woman who works “deftly in the fields” ( ) no longer is able to experience the offerings of life. The “wire cuts,” ( ) pushing her away from the normal flow of life. In addition, man undergoes tragic obstacles as a result of war. “A man walks nobly and alone” ( ) before the horrible effects of war set in on his life causing disruptions. War enters the life of man destroying the bond man shares with his beloved environment ( ).
War slowly begins to strip away the ideals these boy-men once cherished. Their respect for authority is torn away by their disillusionment with their schoolteacher, Kantorek who pushed them to join. This is followed by their brief encounter with Corporal Himmelstoss at boot camp. The contemptible tactics that their superior officer Himmelstoss perpetrates in the name of discipline finally shatters their respect for authority. As the boys, fresh from boot camp, march toward the front for the first time, each one looks over his shoulder at the departing transport truck. They realize that they have now cast aside their lives as schoolboys and they feel the numbing reality of their uncertain futures.

When the war breaks out, this tranquil little town seems like the last place on earth that could produce a team of vicious, violent soldiers. Soon we see Jim thrown into a completely contrasting `world', full of violence and fighting, and the strong dissimilarity between his hometown and this new war-stricken country is emphasised.
