In this lab project, the microbiology students were given 2 unknown bacteria in a mixed broth each broth being numbered. The goal of this project is to determine the species of bacteria in the broth. They had to separate and isolate the bacteria from the mixed broth and ran numerous tests to identify the unknown bacteria. The significance of identifying an unknown bacteria is in a clinical setting. Determining the exact bacteria in order to prescribe the right treatment for the patient. This project is significant for a microbiology students because it gives necessary skills to them for future careers relating to clinical and research work.
Living organisms undergo chemical reactions with the help of unique proteins known as enzymes. Enzymes significantly assist in these processes by accelerating the rate of reaction in order to maintain life in the organism. Without enzymes, an organism would not be able to survive as long, because its chemical reactions would be too slow to prolong life. The properties and functions of enzymes during chemical reactions can help analyze the activity of the specific enzyme catalase, which can be found in bovine liver and yeast. Our hypothesis regarding enzyme activity is that the aspects of biology and environmental factors contribute to the different enzyme activities between bovine liver and yeast.
The purpose of this laboratory is to learn about cultural, morphological, and biochemical characteristics that are used in identifying bacterial isolates. Besides identifying the unknown culture, students also gain an understanding of the process of identification and the techniques and theory behind the process. Experiments such as gram stain, negative stain, endospore and other important tests in identifying unknown bacteria are performed. Various chemical tests were done and the results were carefully determined to identify the unknown bacteria. I was given unknown organism #14, in order to find out what organism I had, I had to perform several different biochemical tests to identify it. Starting with the Gram stain test, which is performed to differentiate Gram-positive and Gram-negative cells. After staining, when observed through the microscope Gram-positive cells are a purple color with thick peptidoglycan cell walls. Gram-negative cells are a pinkish/red color with thinner cell walls. (handout G. s.) My organism was observed to be pinkish rod shaped meaning it is Gram-negative bacteria.
What do bacteria need to grow? For bacteria to grow the most typical thing that they like ate a warm and moist environment, but that is not all that they like. Bacteria also like and environment with a PH that is normal or close to a human PH and bacteria also like an oxygen rich environment. The places that could be common to find bacteria in a building are a keyboard, a water fountain, and restrooms. A keyboard is a common place for bacteria because it is being touched constantly with hands when people type and hands are warm, so bacteria like them. The water fountain is another place that is common for bacteria to grow because people's warm hands are touching it and also it has water, which causes it to be moist. The last place that bacteria will we commonly found in buildings are restrooms. The bacteria like restrooms because many people are in then and also there is a lot of water in them.

During her presentation, Bonnie Bassler expends many different aspects of communication in a bacterial level and how successful communication is important to survival and efficient functioning. It is evident from her presentation that without a common language to communicate with, bacteria could not work as efficiently as they do, neither to immunize people nor create havoc in them. In her presentation, Bassler asserts the importance of the use of language in communication in bacteria, using rhetorical appeals – logical, ethical and emotional – and how it is consequently related to humans.
After completing my Hot Zone Analysis, I saw several problem areas throughout my schedule. One of the more difficult Hot Zone areas I have is that both ENC 1101 and MATH 0022C have quizzes on mostly the same days. I also had scheduled two online finals on the same day and several quizzes the week before them. This schedule would mean that I would have very little time to review before each one. Another big Hot Zone was my commitment to my church and attending all services offered; this cut back on my time able to study throughout the week.
Two unknown bacteria were provided that were labeled “12A” and “12B”. Both bacteria were then used individually following aseptic technique to create heat-fixed smears. Both bacteria were strategically placed on the slide in order to prevent cross contamination and inaccurate results. A total of three slides were made in order to ensure accuracy. The heat-fixed slides were then Gram stained using the procedure listed in the manual; followed by observation under a Leica Dm500 microscope. The slides were observed under 4X,10X,40X and oil immersion and both unknown reactions were double checked by Dr. McLaughlin.

In our Biology Lab we did a laboratory experiment on fermentation, alcohol fermentation to be exact. Alcohol fermentation is a type of fermentation that produces the alcohol ethanol and CO2. In the experiment we estimated the rate of alcohol fermentation by measuring the rate of CO2 production. Both glycolysis and fermentation consist of a series of chemical reactions, each of which is catalyzed by a specific enzyme. Two of the tables substituted some of the solution glucose for two different types of solutions. They are as followed, Table #5 substituted glucose for sucrose and Table #6 substituted the glucose for pH4. The equation for alcohol fermentation consists of 6 Carbons 12 Hydrogens 6 Oxygen to produce 2 pyruvates plus 2 ATP then finally the final reaction will be 2 CO2 plus Ethanol. In the class our controlled numbers were at Table #1; their table had 15 mL Glucose, 10 mL RO water, and 10 mL of yeast which then they placed in an incubator at 37 degrees Celsius. We each then measured our own table’s fermentation flasks every 15 mins for an hour to compare to Table #1’s controlled numbers. At

Microbes are microscopic life forms, usually too small to be seen by the naked eye. Although many microbes are single-celled, there are also numerous multi-cellular organisms. The human body has 10-100 trillion microbes living on it, making it one giant super-organism. Since the first link between microbes and diseases was made, people have been advised to wash their hands. Scientists, however, have recently started to investigate more closely how the microbes that call the human body home affect our health. While some microbes cause disease, others are more beneficial, working with our bodies in many subtle ways.
Most of my trips were early in the morning before classes or in the afternoon after classes, depending of my schedule.
